Overview

This is a four-drug combination consisting of leucovorin (also known as folinic acid or calcium folinate), fluorouracil (5-fluorouracil, 5-FU), oxaliplatin, and s-adenosyl methionine. The first three drugs—leucovorin, fluorouracil, and oxaliplatin—are commonly used together in the FOLFOX regimen for the treatment of colorectal cancer. Leucovorin enhances the binding of fluorouracil to thymidylate synthase, increasing its cytotoxic effect. Fluorouracil is an antimetabolite that inhibits DNA synthesis by targeting thymidylate synthase. Oxaliplatin is a platinum-based chemotherapeutic agent that forms DNA crosslinks and induces apoptosis in rapidly dividing cells[1][2][3]. S-adenosyl methionine (SAMe) is a naturally occurring compound involved in methyl group transfers; it has been studied as a dietary supplement for various conditions but is not part of standard chemotherapy regimens for cancer.
