LAA peptides (Leukemia-Associated Antigen peptides) are short amino acid sequences derived from proteins overexpressed or mutated in leukemic cells, such as Wilms Tumor 1 (WT1), Preferentially Expressed Antigen in Melanoma (PRAME), and mutated Nucleophosmin 1 (NPM1). These peptides are designed to function as cancer vaccines by being presented on HLA molecules (typically HLA-A2) to stimulate the expansion and activation of antigen-specific cytotoxic T lymphocytes (CTLs). In the treatment of acute myeloid leukemia (AML), LAA peptides are investigated for their ability to target and eliminate leukemic progenitor and stem cells, particularly when used in synergistic combinations with immune checkpoint inhibitors like nivolumab or hypomethylating agents like azacitidine.
