Leukocyte interleukin + cyclophosphamide + indomethacin + zinc is a combination immunotherapy regimen primarily investigated as a neoadjuvant treatment for locally advanced, previously untreated squamous cell carcinoma of the head and neck. The combination consists of leukocyte interleukin (LI, branded as Multikine), a defined mixture of 36 pro-inflammatory human cytokines derived from healthy donor blood which stimulate anti-tumor immune responses, combined with cyclophosphamide (immunomodulatory agent and alkylating chemotherapy), indomethacin (NSAID that reduces prostaglandin-mediated immune suppression), and zinc (important for immune function)[3][2][4]. The regimen aims to activate and direct the immune system against tumor cells while modulating the tumor microenvironment and reducing suppressive immune responses. It is administered via local/regional injection (LI) and corresponding oral/IV dosing of the other agents.
