Overview

Leukotriene B4 is a naturally occurring pro-inflammatory lipid mediator derived from arachidonic acid via the 5-lipoxygenase pathway. It is synthesized in myeloid cells, such as neutrophils and monocytes, through the action of 5-lipoxygenase and leukotriene A4 hydrolase. Leukotriene B4 acts primarily as a potent chemoattractant for neutrophils, monocytes, and eosinophils by binding to specific G protein-coupled receptors (BLT1 and BLT2), mediating chemotaxis and activation of these immune cells. This leads to increased production of reactive oxygen species, release of lysosomal enzymes, and amplification of inflammatory responses. Elevated levels are associated with various inflammatory diseases. Leukotriene B4 has been investigated in clinical trials for conditions such as HIV infections and bronchiectasis but is not an approved pharmaceutical product[1][2][8].
