Leukotriene D4 (LTD4) is a naturally occurring cysteinyl leukotriene and a potent inflammatory mediator. It is produced in the body from arachidonic acid via the lipoxygenase pathway and acts primarily as a bronchoconstrictor agent, inducing contraction of smooth muscle in the airways and vasculature, increasing vascular permeability, promoting edema, eosinophil migration, mucus secretion, and contributing to tissue inflammation. Its effects are mediated through high-affinity binding to the cysteinyl leukotriene receptor 1 (CysLT1), a G-protein-coupled receptor that activates downstream signaling pathways such as phosphatidylinositol-calcium second messenger systems. Leukotriene D4 plays an important role in conditions like asthma and allergic rhinitis by sustaining inflammatory responses[1][2][3][5][6]. It has been used experimentally in diagnostic trials for asthma and allergic rhinitis[1][2].
