Overview

Leuprorelin + flutamide is a **combination therapy** used primarily for the treatment of **advanced prostate cancer**. Leuprorelin is a synthetic analog of gonadotropin-releasing hormone (GnRH agonist) that, with continuous use, downregulates pituitary GnRH receptors, leading to reduced luteinizing hormone (LH), follicle-stimulating hormone (FSH), and ultimately testosterone production, resulting in medical castration. Flutamide is a nonsteroidal antiandrogen that blocks the androgen receptor, inhibiting the action of circulating androgens (including both testicular and adrenal testosterone) at their receptor sites. The primary clinical use of this combination is for **androgen deprivation therapy (ADT)**, where the addition of flutamide to leuprorelin mitigates the initial testosterone surge ("flare") and achieves more complete androgen blockade, which has been shown to prolong progression-free and overall survival in some settings of advanced prostate cancer[1][2].
