Overview

This is a combination therapy consisting of three agents: - **Levamisole**: An immunomodulatory agent originally developed as an antihelminthic, now used off-label for its immune-stimulating properties. - **Cyclosporin A**: A calcineurin inhibitor that suppresses T-cell mediated immune responses by inhibiting interleukin-2 production. - **Glucocorticoids**: Steroid hormones with potent anti-inflammatory and immunosuppressive effects, acting through the glucocorticoid receptor to modulate gene expression. The combination is primarily investigated for use in rare hematological disorders such as severe aplastic anemia and paroxysmal nocturnal hemoglobinuria (PNH), especially in refractory or relapsed cases. It has also been studied in lower-risk myelodysplastic syndromes (MDS) and nephrotic syndrome. The rationale behind this regimen is to leverage multiple mechanisms of immunosuppression and immune modulation to induce remission or improve hematologic parameters in diseases with an immune-mediated pathophysiology[1][2][4][5].
