LEVI-04

LEVI-04 is a first-in-class, intravenously administered p75 neurotrophin receptor fusion protein (p75NTR-Fc) developed for the treatment of osteoarthritis and other pain indications. Its mechanism of action involves inhibition of neurotrophin 3 (NT-3) activity by acting as a TrkC receptor antagonist and scavenging excess neurotrophins present in chronic pain states. By supplementing endogenous p75NTR binding protein, LEVI-04 restores neurotrophin homeostasis without ablation of NGF signaling, thus maintaining the neurotrophin function required for cartilage and bone repair. Clinical trials have shown that LEVI-04 provides significant improvements in pain and physical function in patients with knee osteoarthritis, with a favorable safety profile compared to anti-NGF antibodies[1][4][5][6][8].
