TLZ-16-CL (levo-corydalmine hydrochloride) is an orally active small molecule drug developed by Beijing Taizhou Lize Pharmaceutical Technology for the treatment of substance withdrawal syndrome and drug addiction, specifically targeting methamphetamine and cocaine dependence. Derived as an active alkaloid from *Corydalis yanhusuo*, levo-corydalmine acts as a multi-target agent, exhibiting antagonistic activity against dopamine D1, D2, and D3 receptors, alongside agonistic activity at the 5-HT1A receptor. It also inhibits NF-κB activation and modulates NMDA and metabotropic glutamate (mGlu1/5) receptors. By blocking dopamine receptors in the brain's reward pathway, TLZ-16-CL suppresses the hyperlocomotion and behavioral sensitization induced by stimulants like methamphetamine, thereby preventing relapse. It is currently in Phase 1 clinical development in China.
