Levo-demethyl-benproperine hydrochloride is a small molecule derivative of the antitussive drug benproperine, currently under clinical investigation for the treatment of Parkinsonian syndrome. Developed by the Institute of Toxicology and Pharmacology at the Academy of Military Medical Sciences (AMMS) in China, this compound is the levo-enantiomer of the demethylated metabolite of benproperine. While the parent compound is a non-narcotic cough suppressant, levo-demethyl-benproperine is being repurposed for neurodegenerative applications, likely due to its ability to modulate ion channels such as the intermediate-conductance calcium-activated potassium channel (KCa3.1). It has undergone Phase 1 clinical testing in China to evaluate its safety, tolerability, and pharmacokinetic profile in healthy adults.
