Overview

Levoamphetamine is the levorotatory isomer of amphetamine, a central nervous system stimulant that acts as a sympathomimetic agent. While its dextrorotatory counterpart, dextroamphetamine, is more potent in stimulating dopamine release, levoamphetamine exhibits a stronger relative affinity for the norepinephrine system and has a longer half-life. Cognition Therapeutics developed C105 (levoamphetamine sulfate) as a potential treatment for cognitive impairment associated with multiple sclerosis (MS), aiming to improve processing speed and memory deficits. Its mechanism of action involves the release of catecholamines (norepinephrine and dopamine) from presynaptic terminals and the inhibition of their reuptake, primarily mediated through the activation of the trace amine-associated receptor 1 (TAAR1) and interaction with monoamine transporters.
