Levoclausenamide (also known as L-clausenamide or (-)-clausenamide) is a small molecule pyrrolidinone derivative being developed by Qingdao Huanghai Pharmaceutical for the treatment of Alzheimer's disease. It is the levorotatory enantiomer of clausenamide, a compound originally isolated from the Chinese herb *Clausena lansium*. Levoclausenamide acts as a neuroprotective agent with a multi-targeted mechanism of action; it has been shown to enhance synaptic plasticity and long-term potentiation (LTP), inhibit the hyperphosphorylation of tau protein (potentially via GSK-3β inhibition), and protect neurons against amyloid-beta-induced toxicity. Clinical development is primarily centered in China, where it has progressed into Phase III trials.
