Overview

Levodropropizine is a peripherally acting, non-opioid antitussive (cough suppressant) used to treat and manage cough in both adults and children. It is the S-enantiomer of dropropizine and belongs to the class of phenylpiperazines. Unlike centrally acting antitussives such as codeine or dextromethorphan, levodropropizine acts primarily on peripheral airway sensory nerves. Its mechanism involves modulation of sensory neuropeptide levels within the respiratory tract, particularly by inhibiting the release of neuropeptides from C-fibers, thereby reducing cough reflex sensitivity. Levodropropizine has demonstrated efficacy in reducing cough intensity, frequency, and nocturnal awakenings with a favorable safety profile compared to central antitussives[1][3][5]. It may also have anti-allergic properties through affinity for Histamine H1 receptor[6]. The drug is available in oral forms (tablet and syrup) for short-term use.
