Levodropropizine is a peripheral antitussive that suppresses the cough reflex by inhibiting the release of sensory neuropeptides, such as substance P and calcitonin gene-related peptide (CGRP), from C-fibers in the tracheobronchial tree. Unlike opioid-based antitussives, it lacks significant central nervous system activity, reducing the risk of sedation and respiratory depression. Guaifenesin is an expectorant that works by irritating the gastric mucosa, which triggers a vagal reflex that increases the volume and decreases the viscosity of respiratory tract secretions, thereby facilitating mucus clearance. This combination provides a dual therapeutic approach by suppressing the urge to cough while simultaneously aiding in the expulsion of phlegm.
