NTS02 (also known as NTC015) is a fixed-dose combination otic solution containing the fluoroquinolone antibiotic levofloxacin and the corticosteroid dexamethasone. Developed by the Italian pharmaceutical company NTC (Novel Therapeutic Concepts), it is primarily indicated for the treatment of bacterial acute otitis media (AOM), specifically in pediatric patients with tympanostomy tubes (AOMT). Levofloxacin provides broad-spectrum antibacterial activity by inhibiting bacterial DNA gyrase and topoisomerase IV, which are essential for DNA replication, transcription, and repair. Dexamethasone acts as an anti-inflammatory agent by binding to the glucocorticoid receptor, thereby reducing the production of inflammatory mediators and alleviating ear pain and swelling. NTS02 is part of NTC's "Antibiotic Rationalization" initiative, which aims to optimize antibiotic therapy to improve efficacy and reduce the risk of resistance. The candidate has been evaluated in Phase 3 clinical trials, demonstrating its potential as a topical treatment for ear infections.
