Overview

Levomethamphetamine (also known as R-(-)-methamphetamine or levmetamfetamine) is the levorotatory enantiomer of methamphetamine. Unlike its dextrorotatory counterpart (S-methamphetamine), it possesses significantly lower central nervous system activity and is primarily utilized for its peripheral sympathomimetic effects. It acts as an agonist of the Trace amine-associated receptor 1 (TAAR1) and as a selective norepinephrine releasing agent, leading to vasoconstriction. While it is widely available as an over-the-counter nasal decongestant (e.g., Vicks Vapor Inhaler), it is currently the subject of a Phase 1 clinical trial at the University of Wisconsin-Madison. This study, led by Professor Heather Barkholtz and funded by the National Institute of Justice, aims to characterize the drug's pharmacokinetics, metabolism, and potential for impairment to assist in forensic toxicology and the differentiation of over-the-counter use from illicit methamphetamine consumption in drug testing scenarios.
