Overview

Lexgenleucel-T is an autologous cell-based gene therapy developed by VIRxSYS Corporation for the treatment of HIV-1 infection. The therapy involves the ex vivo genetic modification of a patient's own CD4+ T cells using a lentiviral vector (VRX496). This vector delivers a 937-base pair antisense RNA sequence specifically targeting the HIV-1 envelope (*env*) gene. Once the modified cells are re-infused into the patient, the antisense RNA inhibits the replication of HIV-1 within those cells by interfering with the production of viral envelope proteins. This mechanism is intended to protect the CD4+ T cell population from viral-mediated destruction, thereby preserving immune function and potentially reducing the systemic viral load. Lexgenleucel-T was notable for being one of the first applications of lentiviral vector technology in human clinical trials.
