LFA102 is a humanized monoclonal antibody that targets the prolactin receptor (PRLR). By binding to the extracellular domain of PRLR, it prevents the peptide hormone prolactin from activating its receptor. This antagonism blocks downstream signaling pathways implicated in tumorigenesis and proliferation in certain cancers, particularly breast and prostate cancer. Preclinical studies demonstrated that LFA102 could inhibit PRL-induced signaling and tumor growth in vitro and in vivo. However, clinical trials did not show significant antitumor activity as a single agent. The drug was developed by Novartis for use primarily in patients with advanced or metastatic breast cancer, castration-resistant prostate cancer (CRPC), or uterine leiomyoma[1][2][5][6][8].
