LFB083 is a small molecule drug candidate being developed by Leapfrog Bio that targets a synthetic lethality relationship with PIK3CA gain-of-function mutations. Identified through the OncoSLX platform, which screens shelved or deprioritized pharmaceutical assets, LFB083 is designed to inhibit PIK3CA-mutant driven PI3K signaling. The compound has a clinical history, having been previously tested and shown to be safe in approximately 100 unselected patients. It is currently in the IND-enabling phase of development, with a focus on treating breast cancer and other genetically defined PIK3CA-mutant cancers. The drug is expected to be IND-ready in 2027.
