Overview

LFBBz chimeric antigen receptor T cells are an investigational cell therapy designed to target CD70-expressing tumors, specifically glioblastoma. The construct is a ligand-based CAR, utilizing the natural ligand of CD70, which is CD27 (often the extracellular domain), rather than a traditional single-chain variable fragment (scFv). This ligand-based approach is intended to improve binding specificity and reduce potential immunogenicity. The signaling domain of the CAR includes a 4-1BB (CD137) costimulatory domain and a CD3zeta (CD3z) T-cell activation domain (hence 'BBz'). Preclinical studies in glioblastoma models have demonstrated that ligand-based CD70 CARs may offer superior efficacy and persistence compared to scFv-based alternatives, effectively eliminating tumor cells in both in vitro organoid models and in vivo orthotopic xenografts.
