Overview

LFG316 + intravenous immunoglobulin is a combination of two agents: **LFG316 (tesidolumab)**, a human IgG1 monoclonal antibody targeting complement protein C5, and **intravenous immunoglobulin (IVIG)**, a polyclonal immunoglobulin product purified from human plasma. LFG316 acts by binding to complement component C5, inhibiting its cleavage and thereby blocking formation of pro-inflammatory C5a and membrane attack complex C5b-9; this leads to inhibition of terminal complement–mediated inflammation and cell destruction[1][3]. IVIG is a pooled preparation of human immunoglobulin G used as broad immunomodulation, anti-inflammatory, and anti-autoimmune agent, with multiple mechanisms including Fc receptor blockade, modulation of complement activation, and neutralization of pathogenic autoantibodies. The combination is not a standard marketed product but may be used in clinical trial or compassionate settings targeting severe complement-mediated disorders. The individual components have primarily been studied in conditions such as geographic atrophy, non-infectious uveitis, and paroxysmal nocturnal hemoglobinuria (LFG316)[1][3], and as established supportive therapy in various autoimmune and immunodeficiency syndromes (IVIG). The combined use would theoretically target both terminal complement activation (LFG316) and provide broad immunomodulation (IVIG).
