Drug intelligence / Profile preview
LFGuard is a prophylactic vaccine candidate being developed by PAI Life Sciences, in collaboration with the University of Illinois, for the prevention of lymphatic filariasis (LF). The vaccine consists of a recombinant, tag-free tetravalent fusion protein antigen known as rBmHAXT, which comprises four antigens (BmHsp12.6, BmALT-2, BmTSPLEL, and BmTPX-2) from the parasitic nematode *Brugia malayi*. LFGuard is formulated with a Toll-like receptor 4 (TLR4) agonist adjuvant, such as GLA-on-Alum (AL019) or Al-T4, to stimulate a balanced Th1/Th2 immune response and generate protective antibodies and memory T cells. The vaccine is currently in preclinical development, with cGMP manufacturing and IND-enabling studies underway to support progression to Phase 1 clinical trials.
