Overview

LFR-260 is an orally bioavailable small molecule inhibitor of the **Mucosa-Associated Lymphoid Tissue Lymphoma Translocation Protein 1 (MALT1)** protease, developed by **Novartis**. MALT1 is a key component of the CBM (CARD11-BCL10-MALT1) complex, which mediates signal transduction from the B-cell receptor (BCR) to the **NF-κB** pathway. In many B-cell lymphomas, particularly the activated B-cell-like (ABC) subtype of diffuse large B-cell lymphoma (DLBCL), MALT1 protease activity is constitutively active, driving oncogenic cell survival and proliferation. By inhibiting MALT1, LFR-260 suppresses NF-κB signaling and induces apoptosis in malignant B-cells. It is currently being evaluated in Phase 1/2 clinical trials for various B-cell malignancies, including DLBCL, mantle cell lymphoma, and chronic lymphocytic leukemia, both as a monotherapy and in combination with other agents such as rituximab or venetoclax.
