**LG1-34** is a novel small molecule inhibitor targeting EYA2 (eyes absent homolog 2), a phosphatase involved in the SIX1-EYA2 complex that promotes oncogenesis in certain cancers. It demonstrates potent antiproliferative effects in SHI-1 acute myeloid leukemia (AML) cells, reducing cell viability in a dose-dependent manner (91%, 68%, 46%, 3%, and 2% viability at 0.1 µM, 0.3 µM, 1 µM, 3 µM, and 10 µM, respectively), unlike the structurally related LG1-137 which lacks this activity. Developed in the context of research elucidating novel roles for SIX1 and EYA2 in leukemia, LG1-34 represents a potential targeted therapy disrupting EYA2 phosphatase activity to inhibit cancer cell proliferation.
