Overview

LG474 is a non-steroidal selective androgen receptor modulator (SARM) originally developed by Ligand Pharmaceuticals. It functions as a high-affinity agonist of the androgen receptor (AR), exhibiting significant tissue selectivity by promoting anabolic effects in skeletal muscle and bone while demonstrating reduced androgenic activity in the prostate and other reproductive tissues. This pharmacological profile was intended to address clinical needs in muscle wasting (sarcopenia), cancer cachexia, and osteoporosis, providing the benefits of androgen therapy without the side effects of traditional anabolic steroids, such as prostate hypertrophy or virilization. Preclinical studies of LG474 demonstrated its ability to increase lean body mass and improve bone mineral density in animal models. However, the compound did not progress into late-stage clinical development and is currently considered a discontinued research compound.
