Overview

LG842 is a monoclonal antibody developed by Lexicon Genetics in collaboration with Genentech, designed to target and neutralize the protein LG842, which is identified as Angiopoietin-like protein 4 (ANGPTL4). ANGPTL4 is a secreted protein expressed primarily in adipose tissue, the liver, and the pancreas, where it functions as a potent inhibitor of lipoprotein lipase (LPL), the enzyme responsible for the hydrolysis of triglycerides in lipoproteins. By blocking ANGPTL4, LG842 enhances LPL activity, thereby lowering circulating levels of triglycerides and cholesterol. While preclinical studies demonstrated significant lipid-lowering effects, the development of ANGPTL4 inhibitors has historically faced challenges due to potential safety concerns observed in animal models, and the program was not advanced to late-stage clinical trials.
