LGD-2226 is an investigational selective androgen receptor modulator (SARM) developed for the treatment of muscle wasting and osteoporosis. It is a nonsteroidal, nonaromatizable, highly selective ligand for the androgen receptor (AR), exhibiting potent anabolic effects in muscle and bone tissue with minimal impact on prostate weight and luteinizing hormone levels compared to traditional anabolic steroids. The compound acts as a full agonist at the AR with high affinity (EC50: 0.2 nM; Ki: 1.5 nM), promoting protein-protein interactions distinct from those induced by testosterone or fluoxymesterone. Preclinical studies have demonstrated that LGD-2226 enhances bone strength, increases bone mineral density, stimulates periosteal bone formation, and maintains muscle mass in animal models without significant androgenic side effects[2][4][7][10]. It is orally bioavailable and has been considered for additional indications such as sexual dysfunction[4][7].
