LGTX-101 is a machine-learning derived, trivalent VHH-based bispecific T-cell engager (TCE) targeting Nectin-4 and CD3. Developed by LabGenius Therapeutics using their proprietary EVA discovery engine, the molecule features a 3:1 Nectin-4:CD3 stoichiometry designed to achieve avidity-driven selectivity. This allows LGTX-101 to differentiate between high Nectin-4 expression on tumor cells (such as urothelial, head and neck, and triple-negative breast cancers) and low expression on healthy tissues, potentially reducing on-target, off-tumor toxicities. The drug incorporates LALA mutations to eliminate FcγR binding and is engineered for high affinity to Nectin-4 with low-to-moderate affinity for CD3. Preclinical data demonstrate potent tumor regression and a pharmacokinetic profile supporting a bi-weekly or monthly dosing schedule.
