LHC165 is a benzonapthyridine small molecule that acts as an agonist of Toll-like receptor 7 (TLR7). It is adsorbed to aluminum hydroxide and administered via intratumoral injection. Upon administration, it activates TLR7, leading to immunostimulatory and potential antitumor effects. Mechanistically, activation of TLR7 by LHC165 triggers innate immune responses including the activation of CD8+ T cells and natural killer (NK) cells, inhibition of regulatory T cell suppressive function, and induction of interferon alpha production. These actions promote tumor antigen-specific adaptive immune responses and durable antitumor activity. Developed by Novartis, LHC165 has been investigated in phase I clinical trials as both a single agent and in combination with spartalizumab (an anti-PD-1 antibody) for the treatment of advanced or metastatic solid tumors[1][2][3][4][5][6][8].
