LHC165 + PDR001 is a combination of **LHC165**, an investigational immune agonist administered by intratumoral injection, and **PDR001 (spartalizumab)**, an anti-PD-1 monoclonal antibody administered intravenously. **LHC165** is designed to activate immune cells locally within the tumor microenvironment and is a Toll-like receptor (TLR) agonist, specifically acting on TLR7, aiming to stimulate innate and adaptive immune responses to attack cancer cells. **PDR001 (spartalizumab)** blocks the programmed death-1 (PD-1) receptor on T cells, augmenting anti-tumor immunity by inhibiting a checkpoint that enables tumor immune evasion. The combination was primarily studied for safety, tolerability, and preliminary efficacy in advanced solid tumors, notably cutaneous melanoma and head and neck squamous cell carcinoma, in a Phase I/Ib clinical trial sponsored by Novartis.
