Drug intelligence / Profile preview
LHMD is a biomimetic, bio-inspired nanomedicine designed for the targeted treatment of acute myeloid leukemia (AML). It consists of a hollow manganese dioxide (MnO2) nanocarrier encapsulated with the chemotherapeutic agent doxorubicin (DOX) and camouflaged with a leukemia cell membrane (LCM) coating. The LCM coating provides homologous targeting to AML cells in the bone marrow. Upon cellular internalization, LHMD disassembles in response to high levels of glutathione (GSH) and endosomal acidity, releasing DOX and manganese ions (Mn2+). DOX induces double-stranded DNA breaks and immunogenic cell death (ICD), while the released Mn2+ directly activates the cGAS-STING pathway, boosting innate antitumor immunity. This synergistic chemo-immunotherapy approach has demonstrated potent anti-AML efficacy in preclinical models.
