Overview

LIB-GEMs is a non-targeting aptamer-drug conjugate (APDC) developed as a negative control for the PTK7-targeted conjugate (PTK7-GEMs) in the treatment of bladder cancer. It consists of a random "library" DNA sequence (a sequence not selected for specific binding) conjugated to three molecules of the chemotherapeutic agent gemcitabine via phosphodiester bonds. Synthesized through automated solid-phase DNA synthesis, LIB-GEMs is designed to release gemcitabine upon exposure to intracellular phosphatases. In preclinical studies, it demonstrated significantly lower binding affinity and cytotoxicity toward bladder cancer cells compared to targeted conjugates, serving to confirm that the efficacy of PTK7-GEMs is mediated by specific aptamer-biomarker interactions rather than non-specific uptake of the DNA-drug complex.
