LIC11 is an unmasked interleukin-15 (IL-15) superagonist designed for cancer immunotherapy. It is a recombinant fusion protein consisting of a human IL-15 mutant (N72D) and the IL-15Rα sushi domain (together forming the RLI complex) fused to the C-terminus of a human IgG1 Fc fragment. This configuration enhances the stability and half-life of IL-15 while potently stimulating the expansion and activation of CD8+ T cells and natural killer (NK) cells. In preclinical studies, LIC11 serves as a high-activity benchmark; however, its systemic administration is associated with significant dose-limiting toxicities, such as weight loss and systemic cytokine elevation, due to its constitutive activity and the cytokine sink effect. It has been evaluated in various murine tumor models, including prostate carcinoma, colon carcinoma, and melanoma, to demonstrate potent antitumor efficacy at the cost of systemic safety.
