Lidamycin phosphate (also known as C-1027 or LDM) is a highly potent antitumor antibiotic composed of a nine-membered enediyne chromophore complexed with an apoprotein. It acts primarily as a DNA-damaging agent, inducing both single-strand and double-strand breaks in DNA through the generation of reactive diradical species. Unlike other enediynes, lidamycin does not require external activation to trigger its cytotoxic activity. Its unique ability to induce oxygen-independent interstrand DNA crosslinks makes it particularly promising for targeting hypoxic tumor cells that are resistant to conventional therapies. Lidamycin has demonstrated strong cytotoxicity against various cancer cell lines and can induce apoptosis via caspase activation and cell cycle disruption[2][6][8]. It has also shown antibiotic activity against Gram-positive bacteria but is mainly developed for oncological indications.
