Lifitegrast-keratolytic conjugate is a novel preclinical small molecule developed by Azura Ophthalmics for the treatment of ocular surface disorders, including meibomian gland dysfunction (MGD), blepharitis, and dry eye disease. The compound is a prodrug conjugate consisting of lifitegrast, an established lymphocyte function-associated antigen-1 (LFA-1) antagonist, chemically linked to a keratolytic agent. Upon topical administration to the eyelid margin, the conjugate is designed to be hydrolyzed by local esterases, releasing both active components. This dual-action approach aims to simultaneously reduce ocular surface inflammation and address keratinized obstructions of the meibomian glands.
