Overview

Ligelizumab is a humanized IgG1 monoclonal antibody that targets immunoglobulin E (IgE). It binds specifically to an epitope in the Cε3 domain of IgE, neutralizing serum IgE and inhibiting its interaction with both high-affinity (FcεRI) and low-affinity (CD23/FcεRII) receptors. This blockade prevents the activation of allergic effector cells, reducing hypersensitivity reactions. Ligelizumab has been investigated primarily for chronic spontaneous urticaria (CSU), severe asthma, and food allergies. Compared to omalizumab, another anti-IgE antibody, ligelizumab exhibits higher affinity for free serum IgE and greater potency in blocking the IgE/FcεRI pathway[1][4][5][8]. The drug was developed by Novartis.
