LIGHT-PSMA-CART is an autologous CAR T-cell therapy designed to treat castrate-resistant prostate cancer. It involves lentivirally transduced T cells engineered to express a chimeric antigen receptor (CAR) specific for Prostate-Specific Membrane Antigen (PSMA) found on prostate cancer cells. Additionally, these CAR T cells are co-engineered to express LIGHT (TNFSF14), a tumor necrosis factor superfamily member, which aims to enhance CAR T-cell trafficking and cytotoxicity by reversing the immunosuppressive tumor microenvironment and promoting the formation of tertiary lymphoid structures.
