Ligosan

Ligosan (also known as Ligosan Slow Release) is a locally delivered, biodegradable subgingival gel formulation of doxycycline hyclate (14% w/w doxycycline base) developed by Kulzer (formerly Heraeus Kulzer). It is indicated as an adjunct to conventional non-surgical mechanical periodontal therapy (scaling and root planing) for the treatment of chronic and aggressive periodontitis in adults with pocket depths of 5 mm or greater. The formulation utilizes a polyglycolic acid (PGA) and poly(poly(oxyethylene)-co-DL-lactic acid/glycolic acid) (PEG-PLGA) hydrogel matrix to provide sustained, controlled release of the active antibiotic directly into the periodontal pocket for up to 11 to 12 days. Doxycycline acts primarily as a broad-spectrum tetracycline antibiotic by binding to the bacterial 30S ribosomal subunit and inhibiting protein synthesis. Additionally, it functions as a host modulator by inhibiting matrix metalloproteinases (such as MMP-8 and MMP-9), thereby reducing connective tissue breakdown and inflammation in the periodontal pocket.
