Ligustroflavone is a natural flavonoid compound primarily isolated from the fruit of *Ligustrum lucidum* (Glossy Privet). It is being researched for its potential therapeutic applications in chronic kidney disease (CKD), osteoporosis, and various inflammatory disorders. The compound's primary mechanism of action involves the modulation of the GSK3β/NRF2 signaling axis. By inhibiting GSK3β, ligustroflavone prevents the degradation of NRF2, allowing it to translocate to the nucleus and activate the transcription of antioxidant enzymes such as glutathione peroxidase 4 (GPX4). This pathway effectively inhibits ferroptosis—an iron-dependent form of regulated cell death—thereby protecting renal tubular epithelial cells from oxidative damage and reducing interstitial fibrosis in the kidneys.
