Overview

LIM109 (also known as LIM-109) is a small molecule inhibitor of LIM kinase (LIMK), specifically targeting both LIMK1 and LIMK2 isoforms. Developed by Lexicon Pharmaceuticals, the compound was primarily investigated for the treatment of glaucoma and ocular hypertension. LIM kinases are key regulators of the actin cytoskeleton, acting downstream of Rho-associated kinases (ROCK) to phosphorylate and inactivate cofilin. By inhibiting LIMK, LIM109 promotes actin depolymerization and remodeling within the trabecular meshwork, thereby increasing aqueous humor outflow and reducing intraocular pressure (IOP). While LIM109 served as a significant lead in Lexicon's ophthalmic program, the company later advanced other candidates, such as LX7101, into clinical development.
