Limertinib (ASK120067) is a third-generation epidermal growth factor receptor (EGFR) tyrosine kinase inhibitor (TKI) developed by Jiangsu Aosaikang Pharmaceutical (ASK Pharm) and commercialized in China in collaboration with Innovent Biologics. It is designed to selectively and irreversibly inhibit both EGFR-sensitizing mutations (such as exon 19 deletions and L858R) and the acquired T790M resistance mutation, while sparing wild-type EGFR to minimize off-target toxicities. Limertinib has demonstrated significant clinical efficacy in patients with locally advanced or metastatic non-small cell lung cancer (NSCLC), including robust activity against central nervous system (CNS) metastases. It is approved in China for both the second-line treatment of T790M-mutated NSCLC and the first-line treatment of EGFR-mutated advanced NSCLC.
