LIN28-C1632 is a small molecule inhibitor designed to disrupt the interaction between the RNA-binding protein LIN28 and let-7 microRNA. LIN28 (including isoforms LIN28A and LIN28B) is an oncogenic protein that represses the maturation of the tumor-suppressive let-7 miRNA family by binding to the terminal loop of precursor let-7 (pre-let-7). LIN28-C1632 specifically binds to the cold shock domain (CSD) of LIN28, thereby preventing its association with pre-let-7 and restoring the biogenesis of mature let-7. This restoration leads to the downregulation of oncogenic let-7 targets such as MYC, RAS, and HMGA2. Primarily used as a chemical probe in preclinical research, LIN28-C1632 has been investigated for its potential therapeutic effects in various malignancies, including acute myeloid leukaemia (AML), where it may act synergistically with other pathways like Wnt/beta-catenin.
