Overview

Linagliptin + insulin is a combination therapy used for the management of type 2 diabetes mellitus. Linagliptin is a selective, reversible inhibitor of the enzyme dipeptidyl peptidase-4 (DPP-4), which is responsible for the degradation of incretin hormones such as glucagon-like peptide-1 (GLP-1) and glucose-dependent insulinotropic polypeptide (GIP). By inhibiting DPP-4, linagliptin increases the levels of these hormones, stimulating insulin secretion and decreasing glucagon levels in a glucose-dependent manner. Insulin is a peptide hormone that promotes glucose uptake by peripheral tissues and inhibits hepatic glucose production. The combination is often evaluated in clinical settings, such as the COLOR-REAL study led by Yanbing Li, to assess its effectiveness and safety as an add-on therapy in patients whose glucose levels are inadequately controlled by insulin alone.
