Overview
Linagliptin + pioglitazone is a fixed-dose oral combination of two antihyperglycemic agents developed for the treatment of type 2 diabetes mellitus. Linagliptin is a dipeptidyl peptidase-4 (DPP-4) inhibitor that increases incretin levels (GLP-1 and GIP), enhancing insulin secretion and reducing glucagon release in a glucose-dependent manner. Pioglitazone is a thiazolidinedione that acts as an agonist at peroxisome proliferator-activated receptor gamma (PPARγ), improving insulin sensitivity in adipose tissue, muscle, and liver. The combination targets complementary pathophysiological mechanisms in type 2 diabetes to improve glycemic control[1][3][4][6].
