Linaprazan glurate is an orally administered, small molecule prodrug of linaprazan, developed as a next-generation potassium-competitive acid blocker (P-CAB). It acts by competitively inhibiting the potassium ion activity of the gastric H+/K+-ATPase enzyme, resulting in rapid and sustained suppression of gastric acid secretion. Linaprazan glurate is designed for the treatment of erosive gastroesophageal reflux disease (eGERD), including healing and maintenance therapy, especially in patients with moderate to severe eGERD or those not adequately controlled by proton pump inhibitors. The drug has also been investigated for duodenal ulcer and Helicobacter pylori infection. Compared to earlier P-CABs, it offers a longer plasma half-life and improved pharmacokinetic profile, enabling effective 24-hour pH control with a favorable safety profile[1][3][5][7].
