Overview

LinCART19 is a non-viral, plasmid-free chimeric antigen receptor T (CAR-T) cell therapy that targets the CD19 antigen, a B-lymphocyte cell surface protein highly expressed in B-cell malignancies. Developed using linear DNA amplicons transfected into T cells (via electroporation or similar non-viral methods), LinCART19 aims to overcome limitations of viral-vector-based CAR-T manufacturing, offering a potentially safer, faster, and more cost-effective cell therapy platform. The underlying CAR design is derived from constructs already demonstrating complete remission in ALL in clinical trials in China. LinCART19 is being developed for hematological malignancies, especially B-cell leukemias and lymphomas. Its development involves collaborations between iCell Gene Therapeutics (CAR construct) and Applied DNA Sciences/LineaRx (non-viral DNA manufacturing platform)[1][7][9][11][13].
