Overview

Linclatamig is a humanized IgG1 bispecific T-cell engager (TCE) antibody that targets both lymphocyte antigen 6 family member G6D (LY6G6D) and CD3ε on T cells. By binding to LY6G6D expressed on tumor cells and CD3ε on T cells, linclatamig facilitates the formation of an immunologic synapse that redirects cytotoxic T lymphocytes to kill LY6G6D-positive tumor cells. The drug was developed by Genentech (a Roche company) for the treatment of neoplasms, particularly metastatic colorectal cancer with LY6G6D expression. Clinical development reached Phase 1 but was discontinued after limited efficacy was observed in trials; most patients experienced rapid disease progression and only one patient achieved a confirmed partial response[1][4][10]. Skin toxicity such as dry skin and rash were noted as potential on-target off-tumor effects[10].
