LINGO-1-Fc is a recombinant fusion protein designed as an **antagonist** of LINGO-1 signaling pathways in the central nervous system. It acts by blocking LINGO-1’s interaction with receptor complexes that inhibit axonal outgrowth and myelination, specifically the NgR1/p75/LINGO-1 complex. LINGO-1-Fc promotes axonal sprouting, reduces RhoA activation, increases oligodendrocyte and neuronal survival, and facilitates functional recovery after neural injury. In preclinical models, LINGO-1-Fc has been shown to improve remyelination, neurobehavioral deficits, and neuron survival after spinal cord injury and Parkinson’s disease-related damage. The mechanism involves antagonism of LINGO-1-mediated inhibition of EGFR/Akt signaling and modulation of CNS remyelination pathways[1][5].
