Drug intelligence / Profile preview
Linhaliq (formerly known as Pulmaquin or ARD-3150) is an inhaled antibiotic formulation developed by Aradigm Corporation for the treatment of chronic *Pseudomonas aeruginosa* lung infections, particularly in patients with non-cystic fibrosis bronchiectasis (NCFB) and cystic fibrosis. The drug was developed in two primary forms: a purely liposomal ciprofloxacin formulation (Lipoquin, ARD-3100, or CFI) and a dual-release formulation (Pulmaquin, ARD-3150, or DRCFI) which consists of a mixture of liposome-encapsulated and unencapsulated (free) ciprofloxacin. This dual-release mechanism is designed to provide an immediate peak of the antibiotic in the lungs followed by sustained release over 24 hours, allowing for once-daily dosing while maintaining high local concentrations and minimizing systemic exposure. Linhaliq was evaluated in the Phase 3 ORBIT-3 and ORBIT-4 clinical trials. Despite showing efficacy in reducing pulmonary exacerbations in some populations, the FDA issued a Complete Response Letter in 2018, and the program was subsequently discontinued following Aradigm's liquidation in 2020.
