Linperlisib is a next-generation, highly selective, orally administered small molecule inhibitor of the delta isoform of phosphatidylinositol 3-kinase (PI3Kδ). By selectively binding to and inhibiting PI3Kδ, linperlisib blocks activation of the PI3K/AKT signaling pathway, leading to decreased proliferation and increased apoptosis in tumor cells that overexpress PI3Kδ[1][5][6]. Linperlisib has demonstrated compelling clinical activity and a manageable safety profile in patients with relapsed or refractory follicular lymphoma (FL) who have received at least two prior systemic therapies[3][4]. It is also being investigated for peripheral T-cell lymphoma (PTCL), chronic lymphocytic leukemia/small lymphocytic lymphoma (CLL/SLL), other hematologic malignancies, and solid tumors. Linperlisib was developed by Yingli Pharma and was approved in China for relapsed/refractory FL in November 2022. The drug has received FDA Orphan Drug Designations for FL, CLL/SLL, and T-cell lymphoma[5].
